NU 7026 is a novel and potential inhibitor of the DNA repair enzyme DNA-dependent protein kinase (DNA-PK) with IC50 of 0.23 uM; displays selectivity over other PIKK family enzymes.
NU7026 treatment resulted in significant reduction in the number of H2AX foci after -irradiation in wild-type cells, but had no significant effect in RIC1 cells .NU7026 underwent rapid plasma clearance (0.108 l h(-1)) and this was largely attributed to extensive metabolism. Bioavailability following interperitoneal (i.p.) and p.o. administration at 20 mg kg(-1) was 20 and 15%, respectively .
